Trained proofreaders develop an eye for detail, and a good proofreader can spot a mistakenly placed word among other errors. Any incorrectly-sized punctuation marks such as periods and/or commas will not escape the notice of a seasoned and persistent proofreader.
Proofreading is a developed skill, acquired over time. Most errors in written work are made innocently, and detecting errors of clarity and accuracy in one's own work is always difficult. A skilled proofreader reads any text more than twice, very carefully. Sometimes proofreaders read aloud with others, for example, with inspirational writing. This is called a preliminary read; sometimes proofreaders read recently corrected document against an original version of the document: this is called comparative proofreading. Sometimes proofreaders read documents blindly, that is, there are no other documents to compare it to: this is called cold reading.
Proofreaders who proof inspirational writing also pay special attention to Biblical references and historical accuracy that could prevent you from ending up with a real problems of flow and continuity.
Many people find it difficult to spot mistakes when reading text on a computer screen, therefore professional proofreaders read the final copy after printing it out on paper as well. It is intense but rewarding.
